Epithelial Barrier Integrity Assay

The epithelial barrier integrity assay is an in vitro method used to evaluate the integrity of an epithelial cell layer. It plays a crucial role in the safety profiling of drugs and chemicals by assessing their potential to compromise epithelial barrier function.

Purpose of the Test

The epithelial barrier is a key protective physiological barrier of many human organs like the intestine, lungs, and skin. This assay aims to assess the barrier integrity after a specific treatment, by measuring the trans-epithelial electrical resistance (TEER) and/or the passage of marker molecules (e.g. FITC-Dextran) through theepithelium.

The MIVO® Advantage: A Superior Predictive Model

The MIVO® platform revolutionizes the assessment of epithelial barrier integrity by introducing dynamic fluid flow to in vitro tissue models, closely mimicking physiological conditions. MIVO® offers continuous perfusion like the blood circulation, ensuring more accurate and reliable measurements of trans-epithelial electrical resistance (TEER) and marker molecule permeability. This dynamic system allows for a more precise evaluation of how testing substances interact with the epithelial barrier under realistic conditions, providing comprehensive insights into their potential to disrupt or preserve barrier integrity.

Our Unique Value Proposition

Humanized Epithelial Barrier Model: The MIVO® platform integrates a highly relevant in vitro epithelial barrier model that closely replicates the physiological conditions of human tissues, like intestine.
Dynamic Fluid Flow for Enhanced Accuracy: MIVO® employs a unique dual-flow system, simulating both luminal and vascular conditions to better replicate the dynamic environment experienced by intestinal tissue. This ensures a more realistic and precise evaluation of barrier integrity, leading to reliable measurements of TEER and permeability for accurate assessment of substance effects.
Comprehensive and Versatile Testing: The assay is suitable for a wide variety of substances, including pharmaceuticals, nutraceuticals, chemicals, delivering reliable, reproducible results across different industries for comprehensive safety and efficacy evaluations.

In summary

Epithelial barrier integrity assay summary

READOUTS

  • Epithelial barrier integrity (TEER measurement)
  • Tight junctions specific staining
  • FITC-dextran permeability
  • Inflammation (i.e. IL6, IL8)
Epithelial Integrity TEER

TEER measurement is performed before and after exposure to test drug, as indirect measurement of barrier integrity.

Epithelial Integrity FITC

FITC-dextran permeability assay is performed to measure membrane integrity.

Epithelial Integrity IL6

The ability of testing formulation to induce gut inflammation is assessed by quantification of pro-inflammatory cytokines (i.e. IL6, IL8) through ELISA assay.

Epithelial Integrity Tight Junction

ZO-1 immunostaining is performed to investigate the effect of testing compound on tight junctions.
